Ordered differential display of mRNA from <i>Nicotiana tabacum</i>

Abstract
A method that termed ordered differential display of mRNA was applied for tobacco (Nicotiana tabacum L. cv. Havana) for finding differentially expressed genes. RNA was prepared from various explants of tobacco (leaf, non-transformed suspension cell, and human granulocyte-macrophage colony stimulating factor transformed suspension cell). The ordered differential display of mRNA was conducted using those RNAs. Twenty-two extended primer combinations have been used. Among the thirty to sixty distinct bands per each reaction, an average of two to five strong expressed bands was selected. Those bands were further analyzed by DNA sequencing followed by homology search (BLAST). Characterizations of 20 clones were conducted as a preliminary data.
