Polychlorinated biphenyls (PCBs) are a group of persistent organic pollutants with high toxicity and dioxin-like effects. Although globally banned under the Stockholm Convention, PCBs continue to be unintentionally generated from high-temperature processes and chemical manufacturing, leading to potential accumulation in the food chain. This study developed and validated an analytical method for 29 PCBs congeners representing various emission sources, including indicator PCBs, dioxin-like congeners, PCBs characteristic of commercial mixtures, and PCB-209 associated with paint and pigment sources. A QuEChERS–GC-MS/MS procedure was optimized to achieve high extraction efficiency and superior selectivity for seafood matrices, including several species analyzed for the first time in Vietnam. The method was applied to analyze 20 seafood samples. Preliminary results indicated that 2/20 samples contained PCB-28 and PCB-209, while 3 other samples showed traces of certain PCBs at concentrations below the method’s limit of quantitation (LOQ=0.5 µg/kg). Recoveries ranged from 67.0% to 103%, confirming the accuracy and reliability of the method. These findings provided an effective analytical tool to support food safety monitoring and PCBs exposure risk assessment in Vietnam.
