EXPRESSION OF GUS GENE IN INDICA RICE (ORYZA SATIVA SSP. INDICA) DRIVEN BY DlFFIRENT PROMOTER

Abstract

The expression of gus gene (the VidA gene of Escherichia coli that encodes enzyme betaglucuronidase) was studied to analyze the regulation offour promoters including CaMV35S, Rd29A, Gil and Reg2. Results showed that different promoters gave no significant effect on the process ofbiolistics in terms of callus formation and plant regeneration. The temporary expression of gus gene in embryo was observed in all treatments. However, stable expression of gus gene was obser hved in transformed plants when controlled by RCg2 promoter only. Among the transformed (To) plants, expression of gus gene under control of RCg2 promoter was observed in II plants. Results from molecular analysis on these II plants using PCR and Southern blot confirmed the presence of gus gene at 1.1 kb and hph gene at 0.76 kb, which was not observed in non-transformed plants
