Insect Resistance Gene (GNA) Transformation into Cotton by Agrobacterium tumefaciens
Abstract
Hypocotyl explants of cotton (Gossypium hirsutum L.) cv. Coker 312 were transformed with Agrobacterium tumefaciens strain EHA 105 containing the GNA gene under the control of RSs1 promoter and selected on callus initiation medium containing kanamycin. Somatic embryogenesis was promoted in the surviving calli. Plantlets were recovered from putative transgenic embryos. Polymerase Chain Reaction (PCR) and Southern blot analysis were used to confirm the integration of GNA transgene into the plant genome. Western blot result indicated the expressionof the GNA gene in transgenic cotton plants.
