ESTABLISMENT OF HAIRY ROOT LINES IN VIETNAMESE Ficus simplicissima Lour.
Abstract
Ficus simplicissima Lour. contains flavonoids and saponins with strong antioxidant capacity, used to treat a number of diseases such as inflammation, allergies, stomach ulcers... However, the content of natural synthetic flavonoids very low in Ficus simplicissima Lour. (about 0.897 mg/g fresh leaves). Therefore, a method has been proposed to enhance the flavonoid content in Ficus simplicissima Lour. plant is the application of tissue culture techniques to create hairy root lines to increase biomass. This study presents the results of the induction and culture of hairy root lines through A. rhizogenes in Ficus simplicissima Lour. Of the two materials infecting with A. rhizogenes (stem segment, leaf tissue), leaf tissue is the suitable material for hairy root formation. Bacterial density corresponds to OD600 = 0.8; AS concentration 150 μmol/l; infection time 10 minutes; co-cultivation time 2 days; The concentration of cefotaxime 550 mg/l were suitable conditions for induction of hairy root formation from leaf tissue. MS medium in liquid state, without adding growth regulators, cultured in shaking condition is the suitable medium for hairy root growth. The results of checking the presence of rolC gene by PCR method and the absence of virD2 gene confirmed that 5 hairy root lines were created from Ficus simplicissima Lour. plant.
