In this study, NaYF4:Tm3+,Yb3+ upconversion nanoparticles were synthesized via a hydrothermal method
and functionalized with silica, amino groups, and folic acid (FA) for targeted labeling of lung cancer cells.
The nanoparticles exhibited uniform spherical morphology (200–300 nm) and pure hexagonal β-phase,
confirmed by Field emission scanning electron microscopy and X-ray diffraction. Fourier-transform
infrared spectroscopy confirmed the presence of characteristic functional groups, including Si–O–Si
linkages, –NH2, and FA, indicating successful surface modification. Under 980 nm near-infrared excitation,
the functionalized nanoparticles exhibited strong blue emission, with a dominant peak at 475 nm attributed
to the ¹G₄ → ³H₆ transition of Tm³⁺ ions. In vitro fluorescence imaging using Lu-1 lung cancer cells
revealed distinct and selective fluorescence signals, demonstrating the targeting capability of the FA
conjugated nanostructures. These results highlight the potential of NaYF₄:Tm3+,Yb3+@SiO₂-NH-FA as a
promising nanosystem for non-invasive detection and targeted imaging of cancer cells.
