Quantification of gallic acid and caffeic acid in two species of Balanophora J. R. & G. Forst by HPLC

Abstract

With regard the Genus Balanophora J. R. & G. Forst. (Balanophoraceae) contain various phenolic compounds hydozylable tannins, phenolic acids and phenylpropanoids, total polyphenol contents (TPC) of the subspecies Balanophora fungosa and and the variety Balanophora tobiracola Makino, both  recently recorded to the Flora of Vietnam,  the TPC of the species B. fungosa  was determined by Folin-Ciocalteu method. The total polyphenols of B. tobiracola was the highest  (173.22 mg GAE/g). Also, a High performance Thin layer chromatography (HPTLC) procedure was developed and validated for semultaneous quantification of gallic acid and caffeic acid in Balanophora species, as: TLC plate; Mobile phase - toluene-ethyl acetate-formic  acid (14:10:1; v/v/v). Thereby, for the tested samples were specified by biomarkers as: gallic acid - R= 0.34, content = 1.72 - 3.76 mg/g, and caffeic acid - Rf = 0.51; content = 0.24 - 1.32 mg/g. This is the first report on HPTLC analysis of these biomarkers of Balanophora species that may be useful for standardization and quality-control of these species.
