THE EVALUATION OF THE DNA POLYMORPHISM OF SOME RICE LINES REPRODUCED FROM SALT TOLERANT CALLUS

Abstract

Based on the agronomic characteristics, results of biochemical analysis, and assessment of salt tolerance at the germination stage and the 3-leaf seedling stage, we have selected 5 rice lines derived from NaCl 0.1 M salt-tolerant callus originated from CR203. These lines have superiorly different agronomic characteristics in comparison to the original CR203 variety, which is particularly related to rice yields such as panicle length, number of filled spikelets per panicle, and spikelet size. Additionally, their growing time is shorter than the original. These lines are then analyzed their genome by RAPD technique using 10 random primers. The results show that the 10 primers all are polymorphic among 6 rice samples. These results reveal the genetic difference between the selected lines and the original CR203. The genetic difference coefficients range from 0.2406 to 0.4051. Comparing the genetic difference coefficients among the lines shows that the largest difference is between the R3.CR3 line and the R3.CR14 line, at 0.4051. The genetic distance between the selected lines and the original variety is from 0.2785 to 0.3165.
