Abstract

The Asparagus officinalis L. is a high-grade vegetable with rich nutritional content. In addition to being used as an ornamental flower. Research on in vitro propagation of Asparagus in order to provide a uniform, disease-free and cost-effective seed source while meeting the increasing demand for seed is essential. In this study, the factors affecting disease-free prototyping, shoot proliferation, regeneration and shoot growth in Asparagus officinalis L. in vitro culture were investigated. The results showed that sterilization of asparagus seeds with HgCl2 0.1% for 20 minutes gave the best results, the percentage of uninfected germinated seeds reaching 70.83%; Murashige & Skoog medium supplemented with 20 gL-1 sucrose; 2 mg L-1 BAP and 15% v/V coconut water were suitable for shoot growth from seeds (germination rate reached 90% with average height 3.27 ± 0.35 cm after 20 days of culture). A suitable growth promoter for A. officinalis L. shoot multiplication in vitro is the combination of BAP 2 mg L-1 with NAA 0.3 mg L-1. Murashige & Skoog medium supplemented with sucrose 30 L-1; BAP 2 mg L-1 and NAA 0.3 mg L-1 gave the highest shoot multiplication factor, reaching 6.55 times, the shoots are in good quality, big, strong, and dark green.
