Development of an HPLC method for simultaneous determination of quercetin and curcumin I in nanolipid carriers
Abstract
As both quercetin itself and curcumin I are two bioactive compounds commomly loaded in nanostructured lipid carriers (QC-NLCs), an HPLC procedure was developed and validated for simultanous determination quercetin and curcumin I therein (in QC-NLCs). The mothod was established as: Column - Thermo Syncronis RP18 (250 x 4.6 mm; 5 μm; Column temperature - 30oC; Detector - UV (375 nm); Mobile phase – a mixture of acetonitrile and 0.05 % formic acid in water, Flow rate - 1 ml/min; in gradient mode - the ratios of acetonitrile were 47 % and 52 % at 0 - 7 min and 15 - 25 min, respectively. The linearity was good for both quercetin (R2 = 0.9968) and curcumin I (R2 = 0.9962). The % RSD values of intra-day precision of quercetin and curcumin I were 1.80 % and 1.81 %, respectively. The recovery percentages of quercetin and curcumin I were 97.51 – 101.82 % and 97.38 – 102.22 %, respectively. Thus, the proposed method well met the validation requirements and practically usable for establishment of the quality standards of QC-NLCs.
