Development of an HPLC/DAD method for simultaneous quantitative detemination of cleisindosid D and cleistantoxin in the fruits of Cleistanthus tonkinensis (Jabl.) (Euphorbiaceae)
Abstract
An extraction procedure for preparation of the dried extracts from the fruits of Cleistanthus tonkinensis (Jabl.) (Euphorbiaceae) was developed and estimated. Besides, for establishing a product specification of the extracts for medicinal use, an HPLC/DAD method for simultaneous quantification of cleisindosid D and cleistantoxin was developed as: Columns - Inertsil® ODS-3 C18 (250 x 4.6 mm; 5 μm); Mobile phase – CAN-water, in gradient mode. The method was validated regarding cleisindosid D and cleistantoxin for specificity, liniar range (10 % to 250 % of the quantitative concentrations), liniarity (R = 0.9993 and 0.9999); repeatability and reproducibility (RSD < 3.0 %), recovery (95.0 - 105.0 %) and quantitative limit (0.05 and 0.1 µg ml).
