Study on the propagation, growth, and accumulation of kinsenoside compounds of the Anoectochilus roxburghii (wall.) Lindl. orchid under ex vitro conditions

Abstract

Anoectochilus roxburghii (Wall) Lindl. is one of the precious and good herbs for human health. In this study, we investigated the ability of ex vitro propagation, plant growth and the accumulation of the kinsenoside compound of tissue-cultured A. roxburghii (Wall) Lindl. under greenhouse conditions. The results showed that the concentration of NAA ranging from 0 to 1000 ppm were suitable for ex vitro rooting ability and plant growth, with survival rate and rooting explant rate of 100%. The nodes of the plant were capable of regeneration and shoot growth under ex vitro conditions and proved to be suitable materials for ex vitro propagation of A. roxburghii (Wall) Lindl. with 100% shoot regrowth and survival rates. Clean soil substrate of coconut fiber powder was the most suitable medium for transfering tissuecultured plants to outdoor greenhouse conditions, with a plant height of 9.19 cm, root length of 7.11 cm, leaf length of 2.68 cm, leaf width of 1.86 cm, fresh weight of 0.92 g/plant and 100% survival rate. The combination of Nitrophoska fertilizer and Vinh Thanh Biological fertilizer exhibited the best effects on plant growth, with a plant height of 13.29 cm, root length of 11.26 cm, leaf length of 3.54 cm, leaf width of 2.78 cm, fresh weight of 2.62 g/plant and a survival rate of 86.67%. A. roxburghii grew under greenhouse condition at 8 months old showed the ability to accumulate kinsenoside up to 7.19% by dry weight.
