SEQUENCING OF rpoB GENE AND RAPID PROPAGATION OF Lavandula dentata BY TISSUE CULTURE TECHNIQUE

Abstract

In this study, the rpoB gene of L. dentata which was obtained from Da Lat (Lam Dong), was sequencing. It was used to construct the phylogenetic tree. The propagation of L. dentata by tissue culture technique from stem segments was improved. Results showed that the rpoB gene of L. dentata was sequenced, this gene was 483 bp in the length. The analysis of phylogenetic tree expressed that lavender in this work was in the group with L. angustifolia. Stem segments of L. dentata were surface sterilized by 5% javel solution in 10 minutes. The suitable medium for shoots regeneration and multiplication was MS 7 g.L-1 agar, 30 g.L-1 sucrose added 0.7 mg.L-1 BAP. The shoot number per explants was 10.13, the leaf number per shoot was 9.66 and the shoot length was 3.28 cm after 8 weeks of culture. The favorable medium for in vitro rooting was MS, 7 g.L-1 agar, 30 g.L-1 sucrose supplemented 0.5 mg.L-1 NAA. The root number per shoot was 14.66 and the length of root was 1.58 cm after 2 weeks culture. The mixture of soil and coconut fiber (1: 1) was suitable for hardening of in vitro lavender, the survival rate reached 65.0% after 3 weeks of acclimatization.
