ALLELE FREQUENCIES OF 15 AUTOSOMAL STRs LOCI IN MUONG POPULATION LIVING IN HOA BINH PROVINCE OF VIETNAM

Abstract

    In this study, fifteen autosomal STRs loci were analyzed from 104 unrelated healthyindividuals of Muong population living in Hoa Binh Province, Vietnam. The whole bloodsamples were collected from these studied individuals and stored at -20°C. Genomic DNAwas extracted from blood samples using salting out Miller's method. These samples wereamplified simultaneously 15 STR loci (D3S1358, TH01, D21S11, D18S11, PentaE, D5S818,D13S313, D7S820, D16S359, CSF1PO, PentaD, vWA, D8S1179, TPOX and FGA) using amultiplex PCR systems which had been published in our recent research results. The PCRproducts were separated together with Internal Lane Standard 600 (Promega, DG1071) onthe 3130 Genetic Analyzer (Applied Biosystems, Foster City, CA, USA). Data were analysedby GeneMapper® ID software v 3.2.1 (Applied Biosystems, Foster City, CA, USA) accordingto manufacturer's instruction. Allele frequencies and statistical parameters for 15 studied lociwere calculated by EasyDNA_PopuData software which is developed by Wing K. Fung, Yue-Qing Hu and Hong Lee, The University of Hong Kong. The statistical parameters which wereanalysised by this software are observed heterozygosity (OH), expected heterozygosity (EH),standard error of expected heterozygosity (EH_SE), power of discrimination (PD), probabilityof excluding a random man from paternity (PE), chi-square test statistic for the Hardy209Weinberg equilibrium (HWE) of the locus (CHI), 5% critical value of the chi-square test(CHI(c) ) and p-value of the exact test under HWE of the locus (P1). The agreement withHWE was confirmed for 15 studied loci (based on the χ2-test only). The combined power ofdiscrimination (CPD) and the combined power of exclusion (CPE) for the 15 studied lociwere 0,999999999999999998 and 0.999998, respectively.The data in this study can be used for the field of Forensic DNA Casework, DNA Databases,Missing Persons, Parentage and Kinship Testing,....
