EXTRACTION OF COLLAGEN FROM SALMON SKIN (Oncorhynchus mykiss) BY CHEMICAL METHOD
Abstract
The collagen of salmon skin was extracted using acetic acid. The objective of the extraction experiments is the viscosity - measured by viscometer OSVAL. The extraction conditions were optimized by Response Surface Methodology. Firstly, salmon skin was treated with 0.05M NaOH, with a solid/solution ratio of 1/6 (w/v) during 2 hours in order to clean impurities, and then treated in 10% H2O2, with a solid/solution ratio of 1/1 (w/v) in 10 minutes to reduce the pigment in the fish skin. Collagen extraction process was performed with 0.25 M acetic acid, a solid/solution ratio of 1/3 (w/v) in 24 hours. Collagen extracts were precipitated by 4 M NaCl in 5 minutes. The obtained collagen is grey white and in pieces.
