ASSESSING GENETIC HOMOGENEITY OF 10 LINES OF MELON GYNOECIOUS CUCUMBER BY USING SSR TECHNIQUE AND FIELD EXPERIMENT

Abstract

Gynoecious cucumber is a suitable vegetable to cultivate in the net-house. Through the pedigree selection of imported F1 varieties, the inbred lines were selected and made by controlled self-pollination to the seventh generation. The 10 most excellent lines (variety of phenotypes such as rind color, length, weight and productivity) were chosen to assess the homogeneity based on SSR molecular markers and field experiment. 7 SSR markers were used to analyze the purity of 10 lines (10 individuals/line). The result of total DNA extraction showed that the DNA concentration in the range of 101,2-614,8 ng/µl ensured the next steps. Analyzing the genomic homogeneity of 10 lines showed that the homogeneity ranged from 84 to 100%: DL4482 and DL4671 reached 100%; DL4456, 4504, 4412, 4419 and 4442 were over 90%; DL4474, 4399 and 4422 were less than 90%. The weight ranged from 44,0 to 144,3 g; the productivity ranged from 33,8 to 39,0 tons/ha. The analysis of morphological and genetic characteristics showed that some lines with a high degree of purity could be used for crossbreeding
