RESEARCH ON TISSUE CULTURE FOR ARROWROOT (TACCA LEONTOPETALOIDES (L.) KUNTZE) USING APEX BUD

Abstract

Cultured material were 2-week apex buds of Tacca leontopetaloides (L.) Kuntze. Using javel solution (3.5-6.5%) to sterilized the culture samples. Shoot regeneration medium was MS added BA (0.5-2.0mg/l) and NAA (0.1-0.3mg/l) simultaneously. Rooting medium was ½ MS supplemented with IBA (0.0-1.0mg/l) or NAA separately (0.0-1.0mg/l). Research results showed that, sterilized apex bud samples in javel solution at a concentration of 6.5% for 15 minutes gave the best results, the percentage of sterilized samples is 62.2% and all of them are budding; MS medium added BA 1.5mg/l and NAA 0.2mg/l simultaneously gave the highest number of shoots (4.1 shoots/cluster, effective shoots was 3.7 shoots/cluster). Best rooting medium ½ MS associated with IBA 1.0mg/l gave 95.6% rooting shoots, the number of roots was 2.9 roots/bud and the root length was 5cm; The medium consisting of 50% topsoil and 50% coconut fiber gave a survival rate of 69.4% and mean shoot height of 5.6cm after 6 weeks. The research results indicated the prospect for the production of T. leontopetaloides seedlings in-vitro. However, it is necessary to evaluate the adaptability, growth and yield of the tissue seedlings in field before deciding to produce tissue seedlings industrially.
